A popular joint supplement — but the largest independent trial found no significant benefit over placebo for most people with knee osteoarthritis.

What the science says

  • Knee osteoarthritis pain relief Limited evidence

The large NIH-funded GAIT trial found glucosamine (alone or with chondroitin) no better than placebo for knee osteoarthritis pain overall. A possible benefit appeared only in a subgroup with moderate-to-severe pain, and industry-funded trials report better results than independent ones.

  • Slowing cartilage loss Limited evidence

Structural benefits (joint-space preservation) are inconsistent across trials and not established.

Dosage & safety

Studied dose1,500 mg/day of glucosamine sulfate (the form with the most positive data), often combined with chondroitin 1,200 mg/day.
SafetyWell tolerated. Derived from shellfish — caution with shellfish allergy. May slightly affect glucose metabolism; monitor if diabetic.

How to take it

TimingTiming does not matter; split doses through the day if it upsets your stomach.
With food? With food, which reduces nausea and indigestion.
Worth knowingGive it a full 8-12 weeks before deciding, and buy glucosamine sulfate — the hydrochloride form is what most of the negative trials used.

Interactions

With medications

  • warfarin

Well-documented increases in INR and bleeding risk when glucosamine (with or without chondroitin) is added. avoid the combination, or have INR checked closely if your doctor agrees to it

  • insulin and other antidiabetic drugs

Glucosamine may slightly affect glucose metabolism, though clinical trials have mostly been reassuring. monitor blood glucose for the first few weeks if you are diabetic

Well-established interactions only — this is not a complete list. Always tell your doctor and pharmacist what you take.

Frequently asked questions

Does glucosamine work for knee arthritis?

For most people, probably not — the evidence is limited. The large NIH-funded GAIT trial found glucosamine no better than placebo for knee osteoarthritis pain, with a possible benefit only in a subgroup with moderate-to-severe pain.

Does glucosamine rebuild cartilage?

That has not been established: structural benefits of glucosamine, such as preserving joint space, are inconsistent across trials, so the evidence is limited.

How much glucosamine per day, and for how long?

The usual dose is 1,500 mg/day of glucosamine sulfate, the form with the most positive data. Give glucosamine a full 8–12 weeks before judging it.

Is glucosamine safe with warfarin?

Avoid glucosamine with warfarin, since the combination has well-documented increases in INR and bleeding risk. Glucosamine is also derived from shellfish, so take care if you have a shellfish allergy.
